Abstract Scope Dental caries is the most chronic health problem in the world, with the WHO estimating 2 billion people suffer with caries on permanent teeth. Though, polymeric restorative materials have replaced amalgam and revolutionized the treatment of dental caries they suffer from poor longevity due to critical failure in the dental adhesive. Poor adhesive infiltration of demineralized collagen leaves collagen fibers exposed and the polymer adhesive-dentin hybrid layer open to bacterial degradation and further demineralization. This study explores a multi-functional peptide-polymer hybrid to mediate remineralization at this vulnerable interface. Multi-functional peptide-polymer hybrid was first assessed for peptide activity and polymerization properties. Next, the peptide-polymer hybrid system was integrated into a collagen platform and characterized via spectroscopic and microscopic techniques for its ability to remineralize collagen in the presence of polymer resin formulations. Proposed peptide-polymer hybrids offer a next generation effective repair and restoration of caries-affected dentin.
