Abstract Scope |
Bone is a common site of metastasis in prostate cancer (PCa) and breast cancer (BrCa) that leads to morbidity in patients. We have developed novel 3D in vitro nanoclay-based scaffolds that serve as testbeds for bone metastatic PCa and BrCa. Rhodiola crenulate (RC), a Tibetan plant extract, shows some effectiveness towards primary-site cancer, but its efficacy in treating bone-metastasized cancer remains unexplored. We report that RC induces apoptosis in bone-metastasized prostate cancer cells cultured on the 3D scaffold by upregulating pro-apoptotic proteins p53 and caspase-9, while sparing healthy bone cells. Similar results are shown for bone-metastatic BrCa wherein reduced cell viability is observed in bone-metastasized BrCa treated with RC. As for PCa, we observe activation of the pro-apoptotic marker p53 at bone metastasis and high expression of anti-apoptotic marker bcl-2 in BrCa bone-metastatic cultures. These studies indicate the potential for phytochemically enriched plant extracts as new therapeutics for bone metastasis. |
