| Abstract Scope |
Osteogenesis imperfecta (OI) is a genetic bone disorder characterized by increased skeletal fragility resulting from defects in collagen synthesis and matrix organization. Although different OI mutations produce distinct changes in bone composition and structure, the mechanisms linking these alterations to fracture resistance remain poorly understood. To investigate how mutation-specific changes in bone quality influence mechanical behavior and fracture processes, a multiscale experimental framework was adopted to characterize bone structure, composition, deformation, and crack propagation across length scales. By integrating these measurements, we establish relationships between matrix composition, local mechanical heterogeneity, and fracture behavior in OI bone. This work provides new insights into mutation-specific pathways of bone fragility and demonstrates the value of multiscale approaches for evaluating bone quality beyond conventional measures of bone mineral density. |
