| Abstract Scope |
A poor response rate (<20%), serious autoimmune adverse effects, and rapidly washed-out immune stimulants are the main concerns in engineering an efficient drug delivery carrier for cancer treatment. This requires daily injections to keep the immunotherapeutic effect intact & this leads to unwanted systemic toxicity. This work reports the crosslinked stable, immune stimulant-loaded microspheres injectable gel for local immunotherapy. Cross-linked hyaluronic acid gel (xHA) was synthesized using HA (5 % w/v) in a basic medium with BDDE. xHA gel was loaded with iohexol, erythrosine & QS-CpG‐STAT3‐ASO, IL-2, and MMAE drugs, respectively. Results showed an enhanced half-life (~ 35 times) and sustained drug release in-vitro for xHA. xHA remains at the injection sites after 2 days in the pig liver (CT +VE) while un-xHA is washed out. Local quantification of IL2 & CpG in xHA gel was estimated by ELISA, fluorescent spectroscopy, and in oncopig liver tumor model. |
