Electron microscopy is becoming a computational, quantitative discipline: fast direct detectors now make algorithms and physical forward models the limiting factor in turning raw 4D-STEM and spectroscopic data into quantitative 3D chemical structure. In this talk I present a coherent forward-and-inverse computational programme for real specimens. On the inverse side, multi-slice electron ptychographic tomography enables sub-Ångström, near-isotropic 3D resolution beyond the depth-of-focus limit, with joint reconstruction and alignment revealing previously unknown atomic structures and light-element signatures. I then show how fusing direct ptychography with dark-field imaging closes the information gap between low and high spatial frequencies, giving gap-free contrast, depth sectioning, and automated aberration correction in near real time.On the forward side, I introduce a differentiable, calibrated multimodal model unifying ADF, core-loss EELS, and 4D-STEM from one elastic substrate. Powered by BiP-PRISM, it scales beyond one million atoms on a single GPU, enabling simulation-informed, ML-assisted electron microscopy. |
