| Abstract Scope |
Osteogenesis imperfecta (OI) is characterized by impaired bone quality and increased fracture risk. While anti-TGF-β therapy has shown promise in severe forms of OI, its effects on the more common mild subtype remain incompletely understood. We use a multiscale approach to evaluate the effects of anti-TGF-β treatment in a Col1a1+/– mouse model of OI. Anti-TGF-β treatment improved trabecular microarchitecture and reduced regions of elevated strain within bone. However, disruption of the characteristic lamellar mechanical organization still persists, indicating incomplete recovery of hierarchical bone structure. Correlation analyses further showed that treatment altered relationships between matrix composition and local mechanical properties without fully restoring wild type-like behavior. These findings demonstrate that improvements in bone quantity and architecture do not necessarily translate to recovery of bone quality at smaller length scales. The study highlights the importance of multiscale assessment for understanding therapeutic responses and evaluating bone quality beyond conventional measures of bone mass. |
