Hydroxyapatite (HAp) powder is used for liquid chromatography adsorbent for biomacromolecules, e.g, proteins, lipids and nucleic acids. It also possibly adsorbs formazan dyes for colorimetry. However, these adsorptions have not been considered in measurement of cell numbers by amounts of deoxyribonucleic acid (DNA) extraction from cell-extracellular matrices constructs or colorimetry. Three-decades ago, the presenter faced trouble with extraction of DNA from bone-nodule containing cell-collagen sponge constructs and resolved it by use of glycine lysis buffer (pH 10.4) to prevent adsorption of positive DNA onto negatively charged wet synthesized HAp nanocrystals. In this presentation, DNA was extracted from MG-63 cells under presence of HAp nanocrystals (HAp/Col bone-like nanocomposite) and evaluated influence of HAp nanocrystals on DNA extraction as well as an effect of lysis buffer. In addition to those, three measurement methods, Hoechst dye, pico green and nanoDrop®, were compared each other. |
