Presentation Title Implantable Nano-porous Resorbable and Non-resorbable Structures for Cancer Drug Delivery and Tissue Regeneration

Abstract Scope This paper presents concepts for the development of implantable nano-porous bio-resorbable (chitosan) and non-resorbable (poly-di-methyl-siloxane) polymer scaffolds for localized cancer drug delivery and tissue regeneration. Porous chitosan and poly-di-methyl-siloxane drug-loaded nano-porous scaffolds are fabricated using a multi-stage process that involves solvent casting, systematic particulate leaching and phase separation-lyophilization. Anti-cancer drugs, prodigiosin and paclitaxel, are then loaded into the polymer scaffolds. The scaffolds architecture, microstructure, mechanical and thermal properties are elucidated before characterizing their drug release profiles. The insights from in vitro studies of cell viability are also presented for human mammary tumor cell lines (MDA MB 231 and MDA MB 468) and a normal breast cell line (MCF 10A). These include measurements of cell viability, and the results from cytotoxic and apoptotic assays. The implications of the results are then discussed for the development of implantable drug-based bio-resorbable and non-resorbable structures for localized cancer drug delivery and breast tissue regeneration.
