Presentation Title Modeling the Mechanics of Cancer Cells on Tissue Engineering Substrates

Abstract Scope Many diseases have mechanobiological manifestations. The metastasis of prostate and breast cancer lead to migration of cancer cells to distant organs, in particular to the bone. In vitro test-beds to study tumor growth at the secondary site has been developed by our research group. Prostate cancer cells and breast cancer cells are seeded on the scaffolds. The test-beds replicate cancer tumors observed in the human body. A detailed finite element model of a cancer cell comprising of the critical elements of a cell including the cytoskeletal elements such as microtubules, intermediate filaments, actin filaments, cell membrane, cytoplasm and the nucleus is included. Simulations of nanoindentation experiments on cells placed on various scaffold substrates to evaluate the role of substrate stiffness on the nanomechanical response of the cell are conducted. The simulations elucidate mechanisms that influence the nanomechanical response of cells and the influence of the cell-substrate interface on cell mechanics.
