Abstract Scope We synthesized multifunctional mesoporous silicate nanostructures (MSNs): Ca-MSN, Sr-doped CaMSNs (Sr-CaMSNs) and novel Sr-MSNs via sol-gel method for bone regeneration and as antimicrobial peptide (AMP) cargo delivery nano-system. MSNs were characterized for cytotoxicity against mesenchymal stem cells as well as chemically and structurally using scanning electron miscroscopy with energy dispersive X-ray spectroscopy and wide-angle x-ray diffractometry. Peptide loading and release from MSNs was assessed using the fluorescent-labelled GL13K AMP. Ca-MSNs and Sr-CaMSNs were 200±50nm in diameter, and Sr-MSNs were 400±50nm. All MSNs had the expected composition, were amorphous, and had similar peptide-loading efficiency (96 ± 1%) with slow release profile. Sr-MSNs and Ca-MSNs released 7% and 4% of loaded AMP in 72h, respectively. MSNs improved cell metabolic activity, which confirmed their good cytocompatibility. In conclusion, Sr-MSNs support cell growth over extended periods and prolonged AMP release showing potential application for bone regeneration with antimicrobial potency.
